In ageing and disease, there is an increase in specific chemical modifications of proteins called glycoxidative post-translational modifications (GOX). Our research focuses on uncovering the mechanisms whereby these protein modifications cause pathology, specifically in bone. Studies include (i) mapping of GOX modifications in human bone matrix; (ii) determination of their effects on protein structure and function; (iii) characterization of global changes in bone matrix proteome related to GOX-modified proteins.
We also focus on development of novel translatable strategies to predict bone pathology based on identification of GOX-modified bone matrix peptides in serum and urine.
In our laboratory and through multiple collaborations we employ mass-spectrometry, high performance liquid chromatography, differential scanning calorimetry, thermogravimetric analysis and other technologies.
